大鼠肝组织中增生卵圆细胞移植裸鼠后的形态观察

将化学诱癌早期产生的大量卵圆细胞增生肝组织,移植于裸鼠皮下,结合G-6-P酶和γ-GT酶组织化学(简称组化)、AFP免疫组化、~3H-dT放射自显影及超微结构形态学方法,动态地观察卵圆细胞的演变过程。实验结果表明,卵圆细胞显示未分化细胞特征,移植后卵圆细胞仍增生活跃,进一步向过渡细胞、小肝细胞及胆管细胞分化。我们认为卵圆细胞代表肝组织中的一种原始的干细胞,在致癌剂的作用下,由于分化异常,可成为肝细胞癌的组织来源之一。

MORPHOLOGICAL OBSERVATIONS ON THE EVOLUTION OF TRANSPLANTED RAT HEPATIC OVAL CELLS IN NUDE MICE

Liver tissue of rat with prominent oval cell proliferation induced by treatment with 3'-methyl-4-dimethyl-aminoazobenzene (3'-Me-DAB) and 2-acetyl-aminonuorene (2-AAF) from rats on a choline-deficent diet was transplanted subcu-taneously into nude mice. The origin and fate of oval cells were studied using yGT, G-6-Pase histochemical, AFP immunochemi-cal, 3H-dT autoradiographical and electron microscopical techniques, Oval cells appear to be "undlfferentiated", and can further differentiate into transitional cells, small hepatocytes and biliary ductal cells. It is concluded that oval cells may represent"stem-cell" of the liver with differenting polypotentiality, Carcinogens may stimulate oval cell proliferation, but after long exposure oval cells may eventually develop the capacity for carcinogen independent growth. Oval cells may bean origin of hepatocellalar carcinoma.
