大鼠实验性脑外伤后核因子κB活化与细胞凋亡的关系研究

摘    要:目的探讨大鼠实验性脑外伤后核因子κB(NF-κB)活化与细胞凋亡的关系。方法大鼠按实验要求分组,造成脑外伤后不同时间取材,以TUNEL法检测细胞凋亡的发生,以EMSA法检测NF-κB的活化。结果脑外伤后大鼠受创脑域内发生明显的细胞凋亡,同时伴有NF-κB的活化,NF-κB特异性抑制剂PDTC可显著增加凋亡的发生。结论 NF-κB具有保护大鼠脑外伤后细胞免于凋亡的作用,综合考虑NF-κB的活化及凋亡的发生,可为法医学判定脑损伤程度及时间提供依据。

Abstract:Objective To explore the relationship between the activation of nuclear factor κB and cell apoptosis after experimentally traumatic brain injury in rats.Methods TUNEL was applied to detect cell apoptosis while EMSA was used to analyze the activation of NF-κB.Results Cell apoptosis were induced by experimentally traumatic brain injury,along with the activation of NF-κB.PDTC,an inhibitor of NF-κB,enhanced the apoptotic effect.Conclusion NF-κB can protect neuron cells from apoptosis induced by traumatic brain injury.Cell apoptosis and NF-κB activation could be helpful to identify the level and time phase of traumatic brain injury.
