太白楤木对成纤维细胞增殖及形态学的影响

摘    要:[目的]探讨太白楤木对成纤维细胞增殖及形态学的影响及抗肝纤维化机制.[方法]采用体外培养的NIH3T3成纤维细胞作为肝星状细胞(HSC)的替代模型,常规培养,秋水仙碱、齐墩果酸及太白楤木中药血清作用于细胞,在光镜及电镜下观察药物血清对NIH3T3细胞形态学的影响;用放射免疫法测定培养上清透明质酸(HA)的生成.[结果]太白楤木、齐墩果酸、秋水仙碱血清组细胞与对照组细胞在形态、数目上差别显著;3者均可抑制细胞增殖(P<0.01);显著抑制HA的合成,降低NIH3T3细胞培养上清中HA水平,与对照组比较差异有统计学意义(P<0.01).[结论]太白楤木中药血清可显著抑制NIH3T3细胞增殖和细胞外HA的合成,在体外具有一定的抗肝纤维化作用.

Abstract:[Objective]To study the effects of Aralia Chinesis L.on the proliferation and morphology of fibroblasts and to explore the mechanisms involved in its protection against liver fibrosis.[Methods] NIH3T3 fibroblasts were cultured in vitro,as the substitutive model of hepatic satellate cells(HSC).The serum of Colchicinum,Oleanolic acid and Aralia Chinesis L was used respectively.The difference of morphology of fibroblasts which were cultured in different serums was observed by the light microscope and electron microscope and the hyaluronic acid(HA) in culture medium were detected by radioimmunoassay.[Results] The difference of the form and number of fibroblasts between medicinal and control group was remarkable.The serums of three kinds of medicines all significantly inhibited the cell proliferation(P<0.01) and the synthesis of HA as well.The content of HA in culture medium decreased,which differed significantly from those in the control group(P<0.01).[Conclusion] The Aralia Chinesis L.can obviously inhibit NIH3T3 fibroblasts proliferation and decrease the synthesis of HA,indicating the action in anti-liver fibrosis in vitro.
