高脂联合动脉内膜机械性损伤法复制动脉粥样硬化动物模型的实验研究

摘    要:目的]探索一种采用球囊损伤联合高脂饮食的方法建立大鼠腹主动脉内膜损伤,诱导大鼠腹主动脉粥样硬化动物模型,旨在为研究者们提供一种更能贴切模拟动脉粥样硬病变的动物模型,为AS发病机制探讨和治疗方法的探索提供与临床病例更为相似的实验对象。方法]SD大鼠20只,随机分为空白对照组和模型组,每组10只,空白对照组正常饲养,不做任何处理。模型组以高脂饲料喂养4周,自由饮水。4周后,行腹主动脉球囊损伤术,术后继续高脂饲料喂养6周。实验结束后对两组大鼠腹主动脉进行病理切片,观察其病理变化。结果]光镜下观察,模型组大鼠腹主动脉可见粥样斑块病理改变。结论]高脂联合动脉内膜机械性损伤方法复制动脉粥样硬化大鼠模型具有方法可靠,可重复性高,更符合临床动脉粥样硬化疾病的发生发展过程等优点。

Abstract:Objective] Explore a balloon damage by joint high fat diet method to set up big lies aortic lining damage,induction of big lies aortic atheroma animal model for researchers to provide a more appropriate simulation artery atherosclerosis hard lesions in the animal models,for AS the pathogenesis treatment exploration discussion and provide and clinical case more similar subjects.Methods] SD rats only 20,randomly divided into blank control group and the model group,and each group only 10,normally bred blank control group,don’t do anything.Model group with high fat feed 4 weeks,free drink.Four weeks later,do abdominal aortic balloon damage the holonursing continue to high fat feed six weeks.After the end of experiment with two groups of big lies on the aorta pathological slices,observe the pathological changes.Result] By light microscopy observation,model group of big rats aortic visible atheromatous plaque pathological changes.Conclusion] High fat joint artery lining mechanical injury methods copy atherosclerosis in the rat model has the method is reliable,high repeatability,more accord with clinical atherosclerotic disease in the development process,etc.
