摘    要:目的了解缺氧缺血时星形胶质细胞分泌基质金属蛋白酶2(MMP-2)的变化情况及1,6-二磷酸果糖(FDP)能否减少缺氧缺血时星形胶质细胞中MMP-2的表达。方法采用免疫激光共聚集的方法检测体外培养的星形胶质细胞在缺氧缺血不同时段和给予FDP干预时MMP-2的表达。结果星形胶质细胞缺氧缺血12h其MMP-2表达明显增高、24h表达减少,给予FDP干预后缺氧缺血12h与正常对照相比无明显变化。结论缺氧缺血早期星形胶质细胞合成MMP-2增多,对细胞有破坏作用,FDP能通过减少缺氧缺血时星形胶质细胞MMP-2的表达而保护细胞。

Abstract:Objective To investigate the change of expression of MMP-2 protein in cultured astrocytes during hypoxic-ischemic damage and the effects of fructose-1,6-disphosphate(FDP)on expression of MMP-2. Methods The expression of MMP-2 protein was detected by confocus of immunofluorescence in cultured astrocytes of different hypoxic-ischemic period of time and management with FDP.Results The MMP-2 protein expression was significantly increased in hypoxia-ischemia 12 h group and was significantly decreased in hypoxia-ischemia 24 h group compared with the control group.The MMP-2 protein expression in added FDP group was no significant difference after hypoxia-ischemia 12 h compared with the control group.Conclusion The increase of MMP-2 protein expression may be related to the damage of cultured astrocytes during hypoxic-ischemic early period.FDP could proctect cultured astrocytes in hypoxic-ischemic period through downregulated MMP-2 protein expression.
