杜鹃花总黄酮的镇痛作用研究

摘    要:目的研究杜鹃花总黄酮(TFR)的镇痛作用及其机制。方法镇痛试验采用小鼠扭体法、小鼠福尔马林试验、小鼠温浴法及小鼠热板法;血清和脑组织匀浆中NO和PGE2含量的测定分别采用Griess法和紫外分光光度法。结果灌胃TFR(100、200 mg/kg)可明显延长小鼠热板舔足反应潜伏期及小鼠热水缩尾时间且明显抑制小鼠福尔马林试验Ⅱ相反应。TFR(50、100 mg/kg)可明显抑制小鼠扭体反应数;100 mg/kgTFR能升高小鼠血清中NO的含量,200 mg/kg TFR能升高脑组织中NO的含量。TFR可明显降低小鼠脑组织PGE2含量,100 mg/kg可降低血清中PGE2含量。结论TFR具有明显镇痛作用,其镇痛作用机制可能为促进NO释放及抑制PGE2合成。

Abstract:Objective] To study the analgesic effects of total flavone of rhododendron (TFR) and its mechanisms. Methods] By the hot-plate, tail-immersion test,tail-curling reaction and formalin test in mice, the analgesic effects of TFR were observed. Meanwhile, the contents of nitric oxide (NO) and prostaglandin E2 (PGE2) in the mice serum and cerebrum were measured using Griess reaction and spectrophotometry methods, respectively. Results] Lavage with TFR 100,200 mg/kg remarkedly prolonged the latencies of foot-licking as well as inhibited the I phase reaction of mice in the formalin test. 50 and 100 mg/kg TFR inhibited tail-curling reaction in mice. icv TFR 5, 10mg/kg remarkedly reduced the number of the writhing. 100 mg/kgTFR ig significantly increased the mice serum NO content, and 200 mg/kg TFR markedly increased the NO content in mice cerebrum. TFR significantly reduced the contents of PGE2 in mice cerebrum and 200 mg/kg TFR remarkably reduced the PGE2 contents in mice serum. Conclusion] TFR has significant analgesic effect which may be related to increasing the NO contents, reducing the contents of PGE2.
