人结肠癌裸鼠原位移植瘤模型的建立

摘    要:目的 建立人结肠癌裸鼠原位移植瘤模型。方法 使用对数生长期的人结肠癌细胞(HT- 2 9)在裸鼠皮下接种4周形成稳定的皮下移植瘤,再将皮下移植瘤组织块原位接种于2 4只裸鼠盲肠浆膜下,以完成原位移植瘤模型的制备。术后随机分四组,对照组(C组)、塞来昔布(celecoxib)高、中、低剂量组(H、M、L组) ,分别给予纯水及含有不同浓度(15 0 0 ppm、10 0 0ppm和5 0 0 ppm)的塞来昔布的饮水,饲养4 2d后处死裸鼠,观察各组原位移植瘤的成瘤率、瘤体积、重量以及裸鼠实验前后的体重变化,计算抑瘤率。结果 2 4只裸鼠实验期间无一只死亡,成瘤率为10 0 % ,荷瘤鼠实验前后体重变化比较在统计学上无显著差异(P >0 .0 5 ) ,各组原位移植瘤体积P <0 .0 5和瘤重P <0 .0 1比较差异有显著性,L组、M组和H组的抑瘤率分别为2 5 .30 %、38.80 %、76 .92 % ,与对照组比较差异有显著性(P <0 .0 5 )。结论 本实验成功建立了人结肠癌裸鼠原位移植瘤模型,为研究人结肠癌发病机制和干预策略提供了较为理想的动物模型,塞来昔布对人结肠癌原位移植瘤生长具有明显的抑制作用,且有一定的量效关系。

Establishment of Orthotopic Transplantation Model of Human Colonic Tumor in Nude Mice

Abstract:Objects To establish an orthotopic transplantation model of human colonic tumor in nude mice. Methods The cell line HT-29 in log phase of human colonic cancer was implanted subcutaneously in nude mice to develop implant-tumor,and subcutaneously implanted tumor formed 4 weeks later. Then tumor issue was collected and orthotopically implanted in the cecum subserosa of 24 nude mice. Postoperatively these nude mice were randomly allocated into four groups: the control (C)group and high(H),middle(M) and low(L) dose group of celecoxib. Pure water and the water containing 1500 ppm,1000 ppm,or 500 ppm celecoxib were respe ctively provided to these nude mice.These nude mice were sacrificed 42 days later and the rates of tumor formation was observed. The volume and weight of colonic tumor in situ was measured and the changes of weight of all experimented nude mice were also evaluated. At last the rate of tumor inhibition calculated. Results None of the nude mice died, all nude mice formed in situ mass of colonic tumor. The weights of the experimented mice had never changes significantly, while the statistical difference of the volume and the weight of in situ tumor were significant(P<0.05 and P<0.01). The respective rate of the tumor inhibition of L group, M group and H group were 25.30%,38.80%,76.92%,and the difference compared with the control group was significant (P<0.05). The rate of the tumor inhibition was dose-dependenct. Conclusions The successful establishment of orthotopic transplantation model of human colonic tumor in nude mice can provide the ideal model for the study of human colonic tumor pathogenesis and interventionstrategy. The effects of celecoxib inhibiting the growth of the tumor show the obvious dose-dependent tendency in the experiment.
