雌激素和雷洛昔芬对骨质疏松症模型大鼠胫骨干骺端骨折愈合的影响研究

摘    要:目的:研究雌激素和雷洛昔芬对骨质疏松症模型大鼠胫骨干骺端骨折愈合的影响。方法:取♀大鼠随机分为正常对照组、模型组、假手术组、苯甲酸雌二醇(0.5mg.d-1)组和雷洛昔芬(3.4mg.d-1)组,除正常对照组不行手术、假手术组行假手术外,其余各组大鼠均行卵巢切除术,1个月后检测骨质疏松症建模是否成功,取建模成功大鼠和正常对照组、假手术组大鼠,在麻醉状态下,除正常对照组外其余大鼠行胫骨下干骺端截骨术和板内固定术,每组10只,术后所有大鼠每日喂食马铃薯蛋白和相应药物共20g,喂食5周后对各组大鼠胫骨进行生物力学测试(以刚度为指标)、组织形态学分析(以骨痂面积密度、骨小梁节点数为指标)、骨痂总面积测定。结果:与正常对照组、模型组、假手术组比较,苯甲酸雌二醇组和雷洛昔芬组大鼠胫骨的刚度、骨痂面积密度、骨小梁节点数、骨痂总面积均明显增加(P<0.05)。结论:雷洛昔芬可替代雌激素对骨质疏松症模型大鼠胫骨干骺端骨折愈合产生促进作用。

Effects of Estrogen and Raloxifene on Tibia Metaphyseal Fracture Healing of Osteoporosis Model Rats

Abstract:OBJECTIVE: To investigate the effects of estrogen and raloxifene on tibia metaphyseal fracture healing of ovarioto- my model rats. METHODS: Female rats were randomly divided into normal control group, model group, sham operation group, estradiol benzoate group (0.5 rag. d-1) and raloxifene group (3.4 mg. d-1). Except for normal control group, those groups received ovariotomy to establish osteoporosis model while sham operation group received sham operation. In case of same anaesthesia, rats were given tibia metaphyseal osteotomy and internal fixation, 10 rats in each group. After operation, rats were fed with spud pro- tein and relevant drugs 20 g. Biomechanical tests (rigidity as index), histomorphology analysis (poroma area and density, trabecu- la node number as index) and total poroma area of tibia in each group were conducted 5 weeks later. RESULTS: Compared with normal control group, model group and sham operation group, rigidity of tibia, poroma area and density, trabecula node number and total poroma area increased significantly in estradiol benzoate group and raloxifene group (P〈0.05). CONCLUSION: Raloxi- fene can replace estrogen for improving tibia metaphyseal fracture healing of osteoporosis model rats.
