膈下逐瘀汤对大鼠纤维化肝组织Smad4 mRNA表达的影响

目的观察膈下逐瘀汤对肝纤维化大鼠肝脏组织Smad4 mRNA表达的影响。方法SD大鼠60只随机分为空白对照组、模型组和膈下逐瘀汤组共三组，每组20只，用40％四氯化碳（carbon tetrachloride，CCl4）复制大鼠肝纤维化模型，膈下逐瘀汤干预12周后处死大鼠，采用实时荧光定量PCR法检测大鼠肝组织Smad4 mRNA的表达。结果模型组Smad4 mRNA2^-△△Ct值（4．63士0．86）显著高于正常对照组（1．00士0．13）（P〈0．05），表明模型组Smad4 mRNA表达上调。与模型组比较，膈下逐瘀汤组Smad4 mRNA2^△△Ct值（2．08士0．39）显著降低（P〈0．05），表明Smad4 mRNA表达下调。结论膈下逐瘀汤可能通过下调Smad4 mRNA的表达抑制肝脏细胞外基质异常增生，从而发挥抗肝纤维化作用。

Influence of Gexia Zhuyu soup on Smad4 mRNA expression of big mouse fiber liver organization

Objective To observe the Influence of Gexia Zhuyu Soup on Smad4 mRNA expression of big mouse fiber liver organization. Methods 60 SD big mice were divided into control group, model group and soup group at random, every group of 20 mice, by 40% tetrachloromethane （carbon tetrachlo- ride, CCl4） duplicate big mouse liver fiber model, by Gexia Zhuyu Soup intervening, after 12 weeks, measured expression of organizing Smad4 mRNA of the big mouse＇s liver the diaphragm by adopting the real-time fluoro quantitative PCR law. Results Smad4 mRNA 2^-△△Ct in model group（4.63±0.86）was apparently higher than comparative groups（1.00±0. 13） （P〈0.05）, expression of Smad4 mRNA in model group was up adjusted. Smad4 mRNA 2^-△△Ct in soup group（2.08±0. 39） was apparently decreased （P 〈 0. 05）, the expression of Smad4 mRNA was lowered. Conclusions Gexia Zhuyu Soup can inhibit unusual hyperplasia outside the liver substance cell by reducing the expression of Smad4 mRNA, give play to, resist liver fiber take function and play the function of antl--hepatic fibrosis.
