酸枣仁总黄酮在大鼠尿液和粪便中的代谢与排泄

摘    要:目的:研究酸枣仁总黄酮在大鼠尿液和粪便中的代谢与排泄.方法:将SD大鼠灌胃酸枣仁总黄酮提取物后,于不同时间收集尿液和粪便.经大孔吸附树脂处理尿液和粪便,采用高效液相色谱(HPLC)和高效液相色谱-质谱联用(LC-MS~n)法检测尿液和粪便中的原型药和代谢产物.结果:在尿液和粪便中均检测到总黄酮中的主要化合物酸枣仁黄酮碳苷和6'-阿魏酰酸枣仁黄酮碳苷,并发现一个主要代谢产物酸枣仁黄酮次级碳苷(M_1).结论:酸枣仁总黄酮在大鼠尿液和粪便中主要以原型药酸枣仁黄酮碳苷和6'阿魏酰酸枣仁黄酮碳苷的形式排出,少量以代谢产物的形式如酸枣仁黄酮次级碳苷排泄.

Abstract:Objective: To study the metabolism and excretion of total flavonoids of Semen Ziziphi Spinosae in rat urine and feces. Method: The urine and feces of rats were collected at different times after orally administration of total flavonoids from Semen Ziziphi Spinosae, disposed by HP-20 macroporous resin, and then detected by both HPLC and LC-MSn methods. Result: Two main proto-drugs, spinosin and 6¢¢¢-feruloylspinosin, along with a metabolite, swertish (M1), are detected in both urine and feces of rat. Conclusion: Flavonoids of Semen Ziziphi Spinosae are excreted in urine and feces mainly in form of proto-drug, while little in form of metabolites, such as swertish.
