PCNA、EGFR在糖尿病雄性SD大鼠睾丸组织的表达及相关因素分析

摘    要:目的:研究增殖细胞核抗原(PCNA)及表皮生长因子受体(EGFR)在雄性SD糖尿病(DM)大鼠睾丸组织的表达及影响。方法:30只大鼠随机分为正常组(A组)、糖尿病4周组(B组)、糖尿病8周组(c组)各10只。免疫组化SABC法检测睾丸组织EGFR、PCNA,并检测血糖、胰岛素、糖化血清蛋白、体重、睾丸重量、睾酮以分析之间的关系。结果:睾丸免疫组化显示PCNA免疫阳性细胞呈棕黄色颗粒,主要是精原细胞和初级精母细胞,支持细胞也有着色;EGFR在精母细胞、精子细胞、问质细胞和支持细胞均呈阳性反应。糖尿病大鼠PCNA和EGFR阳性率明显低于正常组大鼠(P〈0.05),而随病程进展没有统计学差异,但与血糖呈负相关;PCNA、EGFR与睾丸重量呈正相关;PCNA与血清睾酮水平呈正相关,EGFR与血清睾酮水平无相关性。结论:PCNA、EGFR在一定程度上影响睾丸组织睾酮产生,从而成为影响DM雄性大鼠生殖功能可能的机制之一。

Abstract:Objective: To study the expression and influence of proliferating cell nuclear antigen (PCNA) and epidermal growth factor receptor (EGFR) in SD male diabetic rats testicular tissue. Methods:30 rats were randomly divided into three groups: 10 rats in normal group,10 diabetic rats of 4 weeks group and 10 diabetic rats of 8 weeks group. EGFR and PCNA in testicular tissue were detected by immunohistochemical SABC. The blood glucose, insulin, insulin resistance index, glycosylated serum protein,body weight ,testis weight and testosterone were determined to analyze the relationships among the three groups. Results: It was showed that the positive products of PCNA were brown granules and located at the nuclear of spermatogonium and primary spermatocyte. Sertoli cells were also stained. EGFR were positive in the spermatocyte, sperm cells, stromal cells and support cells. The positive rate in DM group was significantly lower than that of the control (P〈0.05) ,but no statistical difference was observed between the two diabetic group, and negatively correlated with the blood glucose levels; PCNA, EGFR and testis weight was positively correlated; PCNA and serum testosterone levels were positively correlated; EGFR and serum testosterone levels had no correlation. Conclusion: PCNA and EGFR may influence the testosterone production of testicular tissue to a certain excent , which could be one of mechanisms that affect DM male reproductive function.
