北京地区汉族人群胃癌发生与EBVA73基因外显子区A157154C多态性的相关性研究

摘    要:目的 探讨北京地区EBV A73基因外显子区A157154C多态性与胃癌的相关性。方法 采用病例-对照研究方法,以聚合酶链反应(PCR)联合DNA测序检测29例EBV IgM+胃癌患者和40例同期健康体检者EBV A73基因外显子157154 nt位点的基因型和等位基因频率。结果 胃癌组A157154C C等位基因频率和CC基因型频率明显高于对照组(χ2=8.997,P=0.003; χ2=6.801,P=0.033)。C等位基因携带者患胃癌的风险是A等位基因携带者的2.95倍(OR=2.95,95% CI=1.44~6.04)。胃癌组男性患者C等位基因频率显著高于女性患者(χ2=5.392,P= 0.020)。两组间比较显示A157154C不同基因型与年龄、性别、吸烟、饮酒和临床分期均无相关性。结论 EBV A73基因A157154C CA+CC基因型可能是中国北方地区汉族人群胃癌发病的危险因素。

Abstract:Abstract:Objective To investigated the association of EBV A73 gene exon VB variant A157154C and gastric carcinoma(GC)in Han populationof Beijing Area.Methods In this case control study,genotypes of EBV A73(157154 nt)were determined by polymerase chain reaction combined with DNA sequencing in blood samples of 29 GC cases with EBV IgM+ and 40 age-matched healthy controls.Genotype and allele frequency distributions were determined and analyzed.Results The frequencies of the C allele and CC genotype of A157154C variant were significantly higher in the GC group than that in the control group(χ2=8.997,P=0.003; χ2=6.801,P=0.033).Compared with A allele carriers,C allele carriers of A157154Chad a 2.95-fold risk of developing GC(OR=2.95; 95% CI= 1.44~6.04).The Callele frequency in male patients was significantly higher than that in femaleones within the GC group(χ2=5.392,P= 0.020).There was no significant difference found between the GC and control groups in age,gender,smoking,alcohol intake or clinical stages by stratified analyses.Conclusion Suggested a novel association of EBV A73 gene A157154C SNP withGC in a Han population of Northern China.
