一株具有抗HBc及抗HBs双重反应性的抗HBV单克隆抗体细胞株的选育

以富含Dane颗粒的HBV抗原作免疫原进行研究，选育出一株分泌具有抗HBc及抗HBs双重反应性McAb的杂交瘤细胞株。该细胞株经5次连续克隆化证实其克隆单一性，常规染色体检查证实为双倍体细胞（染色体89.5±3.6条），在ELISA中能与基因重组HBcAg、血源性富含Dane颗粒之HBV抗原、小圆颗粒HBV抗原及HBsAg阳性血清发生反应。对四者的反应效价大致相同，但对HBcAg的反应强度显著高于其它三者。此外还发现该McAb在改良双抗原夹心ELISA中能与桥连方式同时与HBcAg及HBsAg相结合。

Establishment of a Cell Line with Anti-HBV Monoclonal Antibody with Two-fold Immunoreactivities against Both HBsAg and HBcAg

In the present study,a hybrldoma cell line(Ell)was established by fusion of mouse myeloma cells(Sp2/0) with spleen cells from BALB/c mice immunized with HBV antigen purified from human plasma and containing a plenty of HBV Dane particles.The monoclonal antibody(McAb)secreted by Ell cell line has two-fold immunoreactivities against both HBsAg polypeptides(HBcAg),HBV antigen containing a plenty of Dane parti-cles,HBV light round particles(both of them come from human plasma)and HBsAg posi-tive human serum as proved by multiple methods.The titers of the McAb ascites that react-ed with above four HBV antigens were basically same,but its immunoreactivity against HBcAg was stronger than that against other three antigens.The specificity of the McAb im-munoreactivity with above-mentioned HBV antigens has been proved by blocking tests and substitution tests.
