摘    要:目的探讨创伤性脑损伤(TBI)大鼠脑组织周期依赖激酶(CDKs)Spy1的表达及细胞定位。方法 72只SD大鼠均分为九组:空白对照组、假手术组和7个TBI组(建立大鼠脑锐器切割伤模型后12h和1、3、5、7、14和28d共7个时间点组)。提取各组大鼠脑组织蛋白样品10μl,Westernblot和逆转录PCR的方法检测TBI后Spy1蛋白和Spy1mRNA表达。免疫荧光双标染色方法检测Spy1蛋白在脑组织中的细胞定位。结果 TBI后,Spy1蛋白及其mRNA的表达逐步升高,3d达高峰,此后逐渐降低恢复至接近正常水平。Spy1主要与星形胶质细胞和神经元之间存在共定位。结论大鼠TBI可以改变脑Spy1的表达,这种改变可能参与脑损伤后星形胶质细胞的活化和增殖过程。

Expression and significance of Spy1 in rats brain tissues after traumatic brain injury

Abstract:Objective To investigate the expression and significance of Spy1(a cycle-dependent kinase) in rats after traumatic brain injury(TBI).Methods Seventy-two SD rats were equally dividedinto 9 groups of blank control(BC),sham operated(SO) and 7 time points after TBI(at 12 h,1,3,5,7,14,28 d after TBI establishment).The expressions of Spy1 protein and its mRNA in bain tissues were detected using Western blot and RT-PCR,respectively.Immunofluorescence was used to observe the distribution of Spy1.Results Compared to group SO,the expressions of Spy1 protein and its mRNA in bain tissues were increased after TBI and peaked on the 3rd day,which were gradually coming down to the normal level thereafter.Double immunofluorescent staining showed that Spy1 co-localized with the markers of astrocyte and neuron.Conclusion The expression of Spy1 may be changed by TBI,which may be involved in activation and proliferation of the astrocytes after brain injury.
