摘    要:目的建立9L/F344大鼠颅内脑胶质瘤模型,用增强MRI动态观察肿瘤生长。方法应用大鼠立体定向仪,在F344大鼠右侧尾状核区接种9L胶质瘤细胞1×105个;观察大鼠的生存状态、体重及活动等情况;分别于大鼠接种后第8、12、17、20、23天在小动物线圈下行大鼠头部强化MRI检查,动态观察肿瘤的生长情况;第20天,取肿瘤标本做组织病理学和神经胶质细胞纤维酸性蛋白(GFAP)免疫组化检查。结果大鼠平均生存期25d。大鼠成瘤率100%。增强MRI动态观察显示同一时期内大鼠的肿瘤体积均一,肿瘤的生长速度和形态符合胶质瘤生长特性;肿瘤组织病理学上接近人恶性脑胶质瘤的病理特点。结论该方法建立的9L/F344大鼠脑胶质瘤动物模型稳定性好,移植瘤生长均一,适合用于胶质瘤的基础研究。

Dynamic observation of intracerebral glioma xenografs growth with enhanced MRI in 9L/F344 rats

Abstract:Objective To establish the rat intracranial glioma xenografts model in 9L/Fischer 344(9L/F344) rats and observe dynamically the tumor growth with enhanced MRI.Methods The 9L gliosarcoma cells were injected stereotactically into the right caudate nucleus in 13 F344 rats.Each rat received 1×105cells in a volume of 10 μl.The rat survival and body weight were observed.The growth of intracranial glioma xenografts was examined dynamically with enhanced MRI on the 8th,12th,17th,20th,and 23rd day after implantation.On 20th day after implantation,3 rats were sacrificed and the tumors were examined with histopathology and GFAP immunohistochemical stain.ResultsThe average survival period of rats was 25 days.The xenograft formation rate was 100%.Enhanced MRI showed that the volumes of xenografts were uniform in the same period and the growth of xenografts was stable.Histopathology features of xenografts were close to those of human malignant glioma sarcoma.Conclusion The 9L/F344 rat intracerebral glioma model has been successfully established,which is growing uniformly and stably,and is suitable for the study on the biological characteristics and therapy of glioma.
