中药抑抗汤含药血清对子宫内膜容受性的影响

摘    要:目的探讨中药抑抗汤含药血清对体外培养的小鼠子宫内膜容受性的影响。方法40只实验小鼠随机分为两组,分别用中药抑抗汤0.8ml和等量生理盐水灌胃28d,制备含药血清和空白血清。提取妊娠第4天(D4)雌鼠子宫内膜细胞后分为含药血清(A组)、胎牛血清(B组)和空白血清(C组)三组进行细胞培养。黏附实验检测子宫内膜细胞的黏附能力,ELISA法检测细胞培养液白血病抑制因子(LIF)水平,Western blot检测整合素ανβ3蛋白水平。结果 A组细胞黏附能力、LIF和整合素ανβ3蛋白表达均较B、C组显著增加(P<0.05)。结论抑抗汤可能是通过增强细胞黏附功能,增加LIF和整合素ανβ3的表达来改善子宫内膜容受性。

Abstract:Objective To investigate the effect of traditional Chinese medicine YiKang decoction(YKD) on the endometrial receptivity.Methods Fourty mice of 4-5 weeks age were eqully randomized into two groups,which were given gastric gavage of YKD 0.8 ml and normal saline 0.8 ml,respectively,for 28 days.The mice serum of two groups was collected.Endometrial cells of pregnant mice were incubated with the serum containing YKD(group A),fetal bovine serum(group B) or the blank mice serum(group C).Adhesion assay was uesd to test adhesion ability of endometrial cells.The leukaemia inhibitory factor(LIF) in the culture mediums was meaured by ELISA.The expression of integrin ανβ3 protein was analyzed by Western blot.Results Cell adhesion ability,LIF and integrin ανβ3 protein levels were higher in group A than those in groups of B and C(P<0.05).Conclusion YKD can improve the endometrial receptivity by enhancing cell adhesion function and increasing the expression of LIF and integrin ανβ3 in the endometrium.
