ZNF267在恶性黑色素瘤中的表达及其对细胞增殖、凋亡和周期的影响

摘    要:目的 研究锌指蛋白267(zinc finger 267,ZNF267)在恶性黑色素瘤组织及细胞中的表达,以及ZNF267对恶性黑色素瘤细胞增殖、凋亡和转移的影响。方法 应用荧光实时定量聚合酶链反应(quantitative real-time polymerase chain reaction,qRT-PCR)方法检测恶性黑色素瘤组织及细胞中ZNF267的表达水平。沉默恶性黑色素瘤细胞中ZNF267的表达后,应用细胞增殖试剂盒-8(cell counting kit-8,CCK-8)检测ZNF267对恶性黑色素瘤细胞增殖的影响,应用流式细胞术检测ZNF267对恶性黑色素瘤细胞凋亡及周期的影响。结果 ZNF267在恶性黑色素瘤组织及细胞中表达水平显著高于癌旁组织及表皮黑素细胞。沉默恶性黑色素瘤细胞中ZNF267的表达后,细胞增殖受到明显抑制,凋亡细胞率显著升高,细胞周期被阻滞于G1期。结论 ZNF267在恶性黑色素瘤中表达显著升高并可促进恶性黑色素瘤细胞的增殖并抑制其凋亡。

Abstract:Objective To investigate the expression of ZNF267 in malignant melanoma tissues and cells,and further investigate the effects of ZNF267 on cell proliferation, cell apoptosis, and cell cycle of malignant melanoma cells. Methods We detected the expression of ZNF267 in tissues and cells of malignant melanoma by qRT-PCR. After silencing the expression of ZNF267 in malignant melanoma cells, we detected the effects of ZNF267 on cell proliferation by CCK-8, we also detected the effects of ZNF267 on cell apoptosis and cell cycle by flow cytometry. Results The expression of ZNF267 was significantly higher in the tissues and cells of malignant melanoma than adjacent tissues and human melanocyte cells. After silencing the expression of ZNF267 in malignant melanoma cells, the cell proliferation was significantly inhibited, cell apoptosis was significantly promoted, and cell cycle was arrested as G1 phase. Conclusion ZNF267 was upregulated in the tissues and cells of malignant melanoma, and could promote the cell proliferation and inhibit cell apoptosis of malignant melanoma.
