摘    要:目的:比较有机酚法和Chelex-100法提取不同组织微量DNA的效果。方法:分别用有机酚法和chelex-100法从外周血、脐带血、绒毛、羊水、胎盘、血斑、口腔分泌物、发根毛囊中提取微量基因组DNA,进行D12S391、D5S818基因座扩增,聚丙烯酰胺凝胶电泳检测分析。结果:外周血、脐带血、绒毛、胎盘、口腔分泌物、发根毛囊均适于用Chelex-100法提取DNA,效果与有机酚法相似。结论:Chelex法是一种高效而快速的微量DNA提取方法,适用于产前诊断和法医鉴定微量组织的检测。

Abstract:Aim: To compare the effect of phenol/chloroform method and Chelex-100 method on micro DNA extraction using different tissues. Methods: The genome DNA was extracted by phenol/chloroform method and Chelex-100 method from peripheral blood, fetal umbilical cord blood, villi, amniotic fluid, placental tissue hair, blood blot, hair, and oral secretion, respectively. With STR D12S391and D5S818 amplification and electrophoresis on denaturation polyacrylamid gel, the extracted DNA was analyzed. Results: The Chelex-100 DNA extracting method was adept to the different tissues, such as peripheral blood, fetal umbilical cord blood, villi, placental tissue, hair, and oral secretion with similar efficiency to phenol/chloroform method. Conclusion: The Chelex-100 DNA extracting method is efficient and quick, which is useful to forensic identification and prenatal diagnosis.
