同种异体骨脱钙骨基质联合BMP修复兔桡骨骨缺损的实验研究

摘    要:目的:探讨异体脱钙骨基质(DBM)骨泥复合骨形态发生蛋白(BMP)修复家兔桡骨节段性骨缺损的作用。方法:日本大耳白兔54只,雌雄不限,随机分为3组,A组:空白组(缺损区不植入任何材料),B组:对照组(缺损区植入金世植骨灵),C组:实验组(缺损区植入DBM骨泥)。建立家兔单侧桡骨骨缺损模型,采用同种异体骨DBM骨泥复合BMP进行修复,复合骨泥中加入骨胶原作为塑形剂,术后4、8、12周观察新骨生成情况,通过形态学、X线、组织学及计算机图像分析等观察指标,并与对照组及空白组进行比较,客观评价骨泥诱导成骨及骨缺损修复能力。结果:DBM与BMP复合骨泥易根据骨缺损大小塑形,术中操作简单。术后A组骨缺损未获骨性修复,B、C组骨缺损均获得骨性愈合,且新骨面积测定显示B、C组间成骨速度无明显差异(P〉0.05),比A组快(P〈O.01)。结论:异体DBM骨泥复合BMP具有良好的骨修复作用。

Abstract:Objective: To discover the effect of material combined bovine bone morphogenetic protein (bBMP) and allogenic demineralized bone matrix (DBM) putty in repairing rabbit radius defect of fracture. Methods: 54 rabbits radius defect models were made and were divided into three groups in random. Group A with nothing,Group B with reconstituted bone xenograft (RBX, jinshizhiguling), and Group C repared with the DBM and bBMP recombined putty. The new bone formation of each group were compared with others by observations, X-ray, histopathological analysis and computer-based image analysis, wich at the period in 4, 8, 12 weeks after operation , to evaluate the bone repairing effect. Results: The DBM putty could be shapen easily according to the defect in operation. Group A was nonunion after operation, complete bone union of the defect were achieved in Group B and Group C,and the measurement of new bone area demonstrated that bone formation speed had no significant differences between the groups of B and C (P〉0.05),but significant difference compared to group A (P〈0.01). Conclusion: Allogenic DBM putty combined bBMP have a good effect in repairing bone.
