摘    要:目的试图比较简并引物扩增GBV-C/HGVNS3变异较大的区域的作用。方法设计了简并引物和减温(touchdown)PCR扩增程序以对目的基因进行扩增,并对阳性PCR产物进行克隆及测序。结果该方法能够检测基因变异约达21%的区域。结论提示该法是一项有效的了解基因家系的技术。

Abstract:Objective To investigate the application of degenerate primers in amplifying highly variable regions of NS3 in GBV C/HGV. Methods The degenerate primers were used to amplify the highly variable regions in NS3 of GBV C/HGV with touchdown PCR program. The positive PCR products were cloned and sequenced. Results Up to 21% deversity regions of the virus were amplified by this method. Conclusion The degenerate primers can be used for amplification of highly variable regions or for indentification of uncharacterized viruses related to known virus groups.
