骨形成蛋白2在大鼠角膜损伤愈合的定位表达及其作用

目的通过观察大鼠角膜损伤愈合过程中骨形成蛋白2（bone morphogenetic protein2，BMP-2）的表达规律来探讨其在大鼠角膜损伤愈合过程中可能发挥的作用。方法雄性Wistar大鼠35只，随机分为7组，每组5只，动物模型组均取右眼制造角膜针刺损伤模型。于损伤后各时间点（6h、1d、3d、5d、7d、14d）取角膜标本，HE染色行组织病理学检查，免疫组织化学染色观察BMP-2的表达与分布，计算机图像分析系统对结果进行分析。结果大鼠角膜上皮BMP-2的表达于损伤后1d显著增高（P〈0．05），损伤后3d、5d时表达逐渐降低，至损伤后14d时表达恢复正常。结论BMP-2在大鼠角膜各部位均有表达，可能参与大鼠角膜损伤愈合过程。

Expression patterns and effect of bone morphogenetic protein 2(BMP-2) on corneal wound healing in rats

Objective To observe the expression of bone morphogenetic protein 2(BMP-2) during corneal wound healing in rats,and to discuss the effects of BMP-2 on corneal wound healing.Methods 35 male Wistar rats were divided into seven groups,five rats in each group.The experimental groups were made into cornea model by needle puncture at their corneas of right eyes.The corneas were surgically removed at six different time points(6 h,1 d,3 d,5 d,7 d,14 d) after operation.The localization and expression of BMP-2 was determined by immunohistochemical methods and computer image analysis.(Results The) expression of BMP-2 of experimental groups reached the maximum on the 1~(st) day in the injured corneal epithelium after operation and significant difference(P<0.05).Then it decreased gradually on the 3~(rd) day and 5~(th) day.And it reached the normal level on the 14~(th) day.(Conclusion BMP-2) may play a role on corneal wound healing after injury.
