食管癌组织p53基因杂合性缺失的检测

摘    要:目的探讨食管癌p53基因杂合性缺失(LOH)与食管癌发生和发展的关系.方法应用荧光原位杂交技术对食管鳞状细胞癌、癌旁不典型增生组织及正常食管上皮各30例进行p53基因LOH检测.结果p53基因LOH检出率在正常食管上皮、不典型增生组织及鳞癌组织中分别为0,16.7%(5/30)和63.3%(19/30),逐渐增高(P<0.05).结论p53基因杂合性缺失是食管癌发生、发展过程中的重要分子学事件.

Abstract:Aim: To reveal the relationship between loss of heterozygosity(LOH) of p53 gene and the carcinogenesis and progression of esophageal carcinoma. Methods: Thirty specimens of esophageal carcinoma, 30 para carcinoma tissues, and 30 normal esophageal epithelial tissues were examined for LOH of p53 gene by fluorescence in situ hybridization(FISH).Results:LOH of p53 gene positive rate was observed in esophageal carcinoma cells (63.3%,19/30), which was significantly higher than that(16.7%,5/30) in para carcinoma and that(0) in normal esophageal epithelial tissues( P < 0.05). Conclusion: LOH of p53 gene plays an important role in the carcinogenesis and progression of esophageal carcinoma.
