改良法兔VX2肝移植肿瘤模型的建立

目的探讨透视导引下经皮穿刺瘤组织块接种法制作兔VX2肝移植瘤模型,并与开腹组织块接种法进行比较。方法新西兰大白兔30只,随机分成2组,15只/组,分别采用透视导引下经皮穿刺接种瘤组织块及开腹瘤组织块接种法种植于肝脏。于接种后14天,行CT平扫与病理组织学结合评价肿瘤接种成功率及生长情况。结果两组肿瘤接种成功率均为100%。穿刺组和开腹组肿瘤最大直径分别为（1.02±0.11）cm和（1.07±0.13）cm,无统计学差异。开腹种植组种植后感染率及肿瘤坏死率明显高于经皮穿刺组（P〉0.05）。结论透视导引下经皮穿刺瘤组织块接种法建立兔VX2肝癌模型具有方法简单、成功率高、动物损伤小、肿瘤坏死率低等特点,为肝癌临床治疗和相关基础研究提供成熟的大型实验动物模型。

The establishment of retrofit model of VX2 carcinoma in rabbit liver

Objective To evaluate the method of establishing model of VX2 carcinoma in rabbit liver using percutaneous puncture inoculation guided by digital subtraction angiography（DSA） and compare with the method of laparotomy inoculation.Methods A total of 30 New Zealand white rabbits were randomly divided into two groups,15 for each group.VX2 tumor masses were implanted into the liver through Angiotech SKATER percutaneous puncture needle guided by DSA in one group and via laparotomic route in another group.Fourteen days later,routine scan of CT and histopathology were used to indicate the success rate of the animal model and the condition of tumor.Results Maximum diameter and the success rate of transplanted tumor had no significant difference between two groups.The success rate of each group was 100 %.The maximum tumor diameter of percutaneous puncture inoculation and via laparotomic route inoculation was（1.02±0.11）cm and（1.07±0.13）cm separately,they had no significant difference（P〉0.05）.The infection rate and the neoplasm necrosis rate of via laparotomic route inoculation was obviously higher than percutaneous puncture inoculation method（P〈0.05）.Conclusion The DSA guided percutaneous puncture inoculation method for establishing rabbit VX2 liver transplanted model is simple,the method has high success rate,little damage to animal and has few of neoplasm necrosis.It is possible to gain a reliable mature transplanted tumor animal model for the study of liver carcinoma therapy and imaging.
