摘    要:目的 :探讨阿司匹林 (ASA)对胃癌细胞株SGC 790 1作用的相关机理 ,为寻找其新的药物用途奠定理论基础。方法 :用MTT法检测药物对细胞的抑制率 ;用流式细胞仪测定细胞周期 ;用琼脂糖凝胶电泳法检测细胞凋亡 ;3H TdR同位素示踪测定细胞DNA合成。结果 :ASA对SGC 790 1细胞株的细胞作用具有量效和时效依赖关系。ASA对细胞DNA合成有抑制作用 ,并可使SGC 790 1细胞周期中S期及G2 /M期比例升高 ,G1期比例下降 ,呈一定的剂量效应关系。琼脂糖凝胶电泳结果未见典型的阶梯状凋亡条带。结论 :阿司匹林对SGC 790 1细胞株存在着细胞毒作用 ,其细胞毒作用可能与抑制DNA的合成、阻止细胞周期有关系

Abstract:Objective To investigate the mechanism of aspirin on the SGC 7901 and to find its new use.Methods Cell cycle was determined using Flow Cytometry (FCM) analysis,Cytotoxicity assay was tested by MTT method.Apoptosis was determined by gelose electrophoresis.DNA synthesis was tested by 3H TdR isotope incroporation.Results As the time and dose increased,the cytotoxicity of aspirin on SGC 7901 was significantly increased.ASA could inhibit DNA syntheses.FCM data showed that,among phases of cell cycle,the S and G 2/M phase ratio were increased significantly and G 1 phase ratio was inhibited in a dose dependent manner.DNA LADDER of typical apoptosis was not found by gelose electrophoresis.Conclusions Aspirin had cytotoxicity on SGC 7901.It maybe associated with DNA synthesis and the phases of cell cycle.
