摘    要:目的 研究氧化的低密度脂蛋白(oxidized low density lipoprotein OX-LDL)对培养的内皮细胞生长状态的影响及其致凋亡作用。方法 用Cu^2+引发脂质过氧化过程,制备不同氧化程度LDL,应用不同氧化程度和不同浓度的OX-LDL,分别作用于对数生长期的内皮细胞,用四唑盐比色试验(MTT)检测法检测细胞活性,碘化丙啶(PI)染色法及酶联免疫吸附法(ELISA)检测细胞凋亡。结果 MTT细胞活性检测显示,轻度氧化的低密度脂蛋(mm-LDL)对细胞生长有促进作用,mm-LDL进一步氧化,其促生长作用转换为抑制作用,与对照组比较差异显著(P<0.05)。OX-LDL对细胞生长呈浓度依赖性抑制作用,ELISA法、PI染色法检测凋亡显示,OX-LDL诱导内皮细胞发生凋亡(与对照组比较P<0.05)。结论 氧化修饰的低密度脂蛋白对内皮细胞的细胞毒性作用与其氧化程度、作用剂量有关,一定浓度的OX-LDL可以导致培养内皮细胞的生长抑制乃至凋亡。

Abstract:Objective To study the effect of oxidized low density lipoprotein (OX-LDL) on the growing and apopto-sis of cultured endothelium cells. Methods Different extent oxidation LDL was obtained by incubating LDL with Cu2 under different conditions Endothelium cells were incubated with different concentrations of LDL oxidized in varying degree. The growing state of cultured endothelium cells were examined by MTT test and apoptosis index was analyzed by PI staining test and FLISA test. Results Within the given concentration limit .minimally oxidized LDL had a promoting effect on the endothelium cells growth. When the minimally oxidized LDL was further oxidized by added Cu2 (5fimol/L) to the cultured system, the promoting effect converted to inhibiting effect. The inhibiting effect of OX-LDL on the endothelium growth was presented in a dose-dependent manner. ELISA test and PI test showed that OX-LDL can induce endothelium cells apoptosis compared with controll group (,P<0. 05). Conclusion The cytotoxicity of OX-LDL to endothelium cells was related to the oxidation extent and the dosage concentration of OX-LDL. OX-LDL. at given concentration, could result in growth inhibition and even apoptosis of cultured endothelium cells.
