TNF-α对大鼠肺微血管内皮细胞SSeCKS mRNA表达的影响

目的研究肿瘤坏死因子（TNF-α）对培养的大鼠肺微血管内皮细胞（PMVEC）中Src抑制的蛋白激酶C底物（SSeCKS）mRNA表达的影响。方法根据TNF-α刺激时间与浓度的差异，将培养的PMVEC随机分为TNF-a时问刺激组与浓度刺激组，前组以5000U／ml的TNF—d分别与PMVEC作用Oh、0．5h、1．5h、3h、6h、12h；后组分别以0U／ml、200U／ml、1000U／ml、5000U／ml的TNF．d与PMVEC作用3h。运用RT-PCR法分析SSeCKSmRNA的表达变化。结果TNF—α可以时间及浓度依赖的方式上调大鼠PMVEC中SSeCKS mRNA的表达。结论SSeCKS可能参与了TNF—α所致大鼠PMVEC单层通透性损伤过程。

Changes of Src-suppressed C kinase substrate mRNA expression in tumor necrosis factor-αinduced rat pulmonary microvascular endothelial cells

Objective To investigate effect of tumor necrosis factor-α （TNF-α） on the Sre-suppressed C kinase substrate （SSeCKS） mRNA in rat pulmonary microvascular endothelial cell（PMVEC）. Methods Cultured PMVEC were randomly divided into two groups. In time-dependent group, cells were cultured with TNF-a （5000U/ml） for 0h,0. Sh,l. Sh,3h,6h,12h, respectively; in dose-dependent group, cells were cultured with TNF-α （0U/ml ,200U/ml,1000U/ml,5000U/ml ） for 3 h. The expression of SSeCKS mRNA was detected by RT-PCR analysis. Results TNF-α upregulated SSeCKS mRNA expression in a time and concentration dependent manner. Conclusion SSeCKS may be involved in the progress of permeability injury of PMVEC monolayer induced by TNF-α.
