肝泰胶囊对大鼠酒精性肝病的防治作用

目的：探讨肝泰胶囊对大鼠酒精性肝病（ALD）的防治作用及其可能的机制。方法：将40只SD雄性大鼠随机分为正常组10只,采用普通饲料喂养;模型组18只,高脂饲料喂养,同时灌喂酒精性混合液;预防组12只,高脂饲料喂养,同时给予肝泰胶囊和酒精混合液。实验时间为4个月。采用光镜、电镜观察大鼠肝组织病理学改变;M asson染色观察各组大鼠肝纤维化程度;采用放射免疫法测定血清透明质酸（HA）、Ⅲ型前胶原（PCⅢ-）、Ⅳ型胶原（Ⅳ）的含量;用半自动生化仪检测丙氨酸转氨酶（ALT）、天门冬氨酸转氨酶（AST）;分光光度仪检测丙二醛（MDA）、超氧化物歧化酶（SOD）、还原型谷胱甘肽（GSH）、γ-谷氨酰转肽酶（γ-GT）;免疫组化检测α-平滑肌蛋白（α-SMA）抗体的表达。结果：预防组与模型组比：①预防组大鼠肝组织肝纤维化程度明显改善;②肝组织胶原纤维面积明显减少;③血清透明质酸显著降低;④肝功能损伤明显减轻。结论：肝泰胶囊能显著改善酒精性肝病大鼠的肝损伤,提示该药对酒精性肝病有一定的防治作用。

Effect of Gantai capsule on alcohol liver disease in rats

Objective：To investigate the possible mechanism of Gantai capsule on resisting hepatic fibrosis induced by alcohol in rats.Methods： Forty rats were randomly divided into control group,model group and Gantai-treated prophylaxis group.Light and electron microscope were adopted to observe the histopathology of the hepatic tissue.Masson dyeing and PC-Ⅲ,HA,Ⅳ-C in the serum were evaluated to reflect the level of hepatic fibrosis.Meanwhile,blood,lipids,liver MDA,SOD,and GSH were measured and then used immunohistochemistry to measure the expression of α-SMA in liver tissue.Results：Compared with model group,it revealed that Gantai-treated prophylaxis group rats： ① The degree of fibrosis was improved.② Liver collagen fibers markedly was decreased.③ Serum hyaluronic acid and laminin obviously was decreased.④ Liver function was improved.Conclusion：Gantai capsule has an effect on alcohol liver disease and can significantly alleviate the alcohol-induced hepatic fibrosis injury in rats.
