摘    要:目的研究锰对大鼠生精功能的影响,探讨锰对大鼠生精功能抑制作用的机理。方法雄性SD大鼠,体重(120±10)g,随机分为6组:空白对照组,低剂量(15mg/kg MnCl2)和高剂量(30mg/kg MnCl2)组,8只/组。空白对照组给予等容NS,给药途径均为腹腔注射,5d/周,1次/d。空白对照组和MnCl2组分别于第4周末和第6周末处死,观察睾丸形态学改变并检测精子质量相关指标。结果与空白对照组比较,各组生精小管呈不同程度变性,生精细胞和精子数量减少,缺如。高剂量4周组睾丸脏器系数和各染锰组精子数量、活动度显著降低,畸形率显著升高(P<0.01)。染锰剂量相同,6周组与4周组比较,精子数量和精子活动度显著降低(P<0.01)。染锰时间相同,高剂量组与低剂量组比较,精子数量和精子活动度显著降低,畸形率显著升高(P<0.01)。结论染锰15mg/kg 4周即可对雄性大鼠生精功能产生抑制作用。

Abstract:Objective To study the effects of manganese on the spermatogenic function of male rats and to discuss its mechanism. Methods 48 healthy male rats (weighting 120 :t: 10 g) were randomly di- vided into 6 groups (blank control,15 mg/kg MnC12 ,30 mg/kg MnCI~ groups, and each was subdibided 4 w and 6 w group). 8 rats/group. 15 mg/kg MnC12 and 30 mg/kg MnC12 groups were contaminated manganese (MnC12 ~ 4H20) by I. P. for 4 and 6 weeks respectively and blank control groups were injec- ted with NS. All rats were injected once everyday, five times per week. In the 4'h and 6th weekend, their testis were collected. The morpho logical changes of testis and the correlative indexes of sperm were ex- amined. Results Compared with blank control groups, the seminiferous tubules showed degeneration in various degree, the number of spermatid was depleded and no spermatocytes were seen in the tubules, the weight coefficient of testis was decreased in 4 w 30 mg/kg MnC12 group (P 〈 0.01 ) , the quantity and activity of sperm were decreased but the ratio of sperm deformity was increased in every manganese expo- sure groups (P 〈 0.01 ). Among manganese exposure groups of same dose, the quantity and activity of sperm were decreased significantly in 6 w manganese exposure groups than in 4 w groups (P 〈 0.01 ). A- mong manganese exposure groups of same time, the quantity and activity of sperm were decreased and the ratio of sperm deformity was increased in 30 mg/kg manganese exposure groups than it in 15 mg/kg groups (P 〈 O. O1 ). Conclusion 15 mg/kg MnC12 exposure for 4 w may have supress effect on the sper- matogenic function of male rats.
