穿心莲致小鼠嗜多染红细胞和成熟红细胞微核的实验研究

摘    要:目的:动态观察穿心莲水煎液对小鼠嗜多染红细胞和成熟红细胞微核的影响。方法:20只小鼠随机分为对照组和实验组,每组10只,分别连续灌胃蒸馏水和穿心莲水煎液15 d,观察嗜多染红细胞和成熟血细胞,比较细胞微核率的差异。结果:实验组嗜多染红细胞和成熟血细胞微核率呈阳性,微核率≥5‰,但成熟红细胞的微核数量明显比嗜多染红细胞低(P<0.05)。结论:穿心莲对小鼠骨髓细胞具有潜在致染色体断裂效应;外周血涂片是观察成熟红细胞微核简单、有效的方法。

Abstract:Objective: To investigate the effects of water decoction of Andrographis Paniculata Nees on mouse polychromatic erythrocytes and mature erythrocytes through dynamic observation of micronucleus. Methods: Twenty mice were randomly divid- ed into control group and experimental group with 10 in each. The mice in control group and experimental group had been per- formed continuous gastric perfusion with distilled water and water decoction of Andrographis Paniculata Nees for 15 days, respec- tively. Polychromatic erythrocytes and mature ~rythrocytes were subsequently isolated and the appearance rates of micronucleus were compared through microscopic examination. Results: In experimental group, the rates of micronucleus appearance in both polychromatic erythrocytes and mature erythrocytes were 〉~ 5 %0. And the micronucleus rate in mature erythrocytes was signifi- cantly lower than in polychromatic erythrocytes. Conclusion: Andrographis Paniculata Nees has potential chromosome breakage effects on mouse bone marrow cells. The peripheral blood smear is an easy and effective method to detect micronucleus in mature erythrocytes.
