端粒酶活性及PCNA表达与大肠癌相关性的研究

目的:研究端粒酶活性、PCNA在大肠癌中的表达特征,探讨其与大肠癌发生发展的关系。方法:运用PCR-TRAP-ELISA及免疫组化法对30例大肠癌,癌旁组织、正常大肠粘膜及20例大肠腺瘤性息肉组织端粒酶活性、PCNA蛋白的表达进行检测。结果:(1)端粒酶活性及PCNA在大肠癌组织中的表达明显高于癌旁组织、正常粘膜及腺瘤性息肉组织(P<0.05);(2)大肠癌组织端粒酶及PCNA蛋白表达与淋巴结有无癌转移密切相关,伴淋巴有癌转移大肠癌患者端粒酶阳性表达率明显高于无淋巴结癌转移者(P<0.05);(3)端粒酶活性与PCNA表达密切相关。结论:(1)端粒酶的检测不仅有助于大肠癌的诊断和鉴别诊断,而且对判断患者预后具有重要意义;(2)PCNA的过度表达可能是端粒酶激活的重要途径之一。

Relationship between Telomerase Activity and PCNA in Colorectal Carcinoma

Objective:To explore relationship between the expressions of telomerase and PCNA,and to reveal the occurrence and devel-opment of colorectal carcinoma.Methods:Telomerase activity was determined by PCR-TRAP-ELISA and expression of PCNA was measuredwith inmmunohistochemical staining in 30 colorectal carcinomas,tumor-adjacent tissues,normal tissues and 20 adenonma polypus.Results:(1)Telomerase activity and the expression of PCNA in colorectal carcinoma were significantly higher than those in other tissues(P<0.05).(2)The positive rates of telomerase and PCNA were higher in cases with lymphatic matastasis than that without lymphatic matastasis(P<0.05).(3)A significant positive relationship was observed between telomerase activity and the expression of PCNA.Conclusion:(1)The detection oftelomerase activity is not only useful in diagnosis and differential diagnosis of coloretal carcinoma,but also helpful in predicting prognosis ofpatient with colorectal carcinoma.(2)The overexpression of PCNA may be one of important pathways by which telomerase is activated.
