缺血性脑卒中大鼠大脑皮层EphrinB2 信号表达变化的研究

摘    要:目的观察大鼠缺血性脑卒中后皮层中磷酸化EphrinB2 的表达。方法将成年雄性Sprague-Dawley 大鼠随机分为假手术组和模型组,每组12 只。采用线栓法制作大鼠大脑中动脉阻塞(MCAO)缺血再灌注模型。利用Western blotting 和免疫组化SP法检测两组大鼠脑皮层中磷酸化EphrinB2 的表达。结果模型组大鼠大脑皮层的磷酸化EphrinB2 表达量和阳性细胞数目均高于假手术组(P<0.05),并且血管内皮有磷酸化EphrinB2的表达。结论大鼠缺血性脑卒中后EphrinB2 信号通路被激活。

Abstract:Objective To observe the expression of phosphorylated EphrinB2 in brain after focal cerebral ischemia/reperfusion in rats.Methods 24 male Sprague-Dawley rats were randomly divided into sham group (n=12) and model group (n=12). The model group was modeled as middle cerebral artery occlusion and reperfusion with nylon monofilament suture, and then was assessed with Longa's score. The expression of phosphorylated EphrinB2 in cerebral cortex was detected with immunohistochemistry and Western blotting. Results The expression of phosphorylated EphrinB2 and the number of positive cells were significantly higher in the sham group than in the control group (P<0.05). It existed in the vascular endothelium in cerebral cortex. Conclusion EphrinB2 signaling pathway is activated in ischemic stroke.
