原发性开角型青光眼小梁网组织早衰的初步研究

摘    要:摘 要: 【目的】 观察正常小梁网组织和POAG小梁网组织在超微结构和衰老相关β-半乳糖苷酶(SA-β-Gal)表达方面是否存在差异?【方法】 正常人小梁网组织取自广东省眼库的正常人眼球(10例),平均年龄(36.90 ± 10.10)岁;POAG小梁网组织来自中山眼科中心青光眼科行小梁切除术者(10例),平均年龄(39.80 ± 11.64)岁?用透射电镜观察两组小梁网组织超微结构;用衰老相关β-半乳糖苷酶(SA-β-Gal)染色法对两组小梁网组织进行染色,并分析两者之间衰老染色的差异?【结果】 透射电镜发现正常人小梁网组织细胞膜完整,核染色均匀居中, 胞浆中可见形态正常的中心体, 周围散落分布着的线粒体?溶酶体等细胞器;基底膜完整连续;弹性纤维周围绕有薄鞘,胶原纤维呈束状排列?整齐;Schlemm管管腔通畅?而POAG患者的小梁网组织中可见细胞脱落溶解,细胞膜不完整,细胞核固缩或肿胀,线粒体水肿,线粒体嵴扩张;Schlemm管管腔变狭窄;弹性纤维增多且排列紊乱;基底膜增厚?SA-β-Gal染色发现POAG小梁网中的衰老细胞阳性率较正常人小梁网组织高(P < 0.001)?【结论】 POAG小梁网超微结构破坏明显,SA-β-Gal染色增强,在POAG小梁网中可能存在早衰?

Abstract:Abstract:【Objective】 To evaluate the differences in the ultrastructure and Senescence β-Galactosidase of trabecular meshwork between the patients with POAG and normal controls. 【Methods】 Trabecular meshworks from 10 eyes with POAG and 10 normal eyes were taken. Normal human eyes were obtained from the Eye Bank in Guangdong Province. The ages of the normal donors mean as 36.90 ± 10.10 years old. The trabecular meshworks specimens from patients with POAG (39.80 ± 11.64 years old) were obtained by standard trabeculectomy for therapeutic purposes. Transmission electron microscope was used to observe the ultrastructure of the trabecular meshwoks. Senescence β-Galactosidase (SA-β-Gal) staining was used to evaluate the senescence of trabecular meshworks of the patients with POAG and normal controls. 【Results】 Intact membranes, centered and homogeneous nucleus were shown in the trabecular meshwork cells of the normal control. Moreover, normal-shaped centrosome, mitochondria, and lysosome could be seen in the cytoplasm. The basement membranes were unbroken. The elastic fibers were surrounded with thin sheath. The collagen fibrils were ranged in bunch. And the Schlemm canals were open. Compared to trabecular meshworks in normal control, the trabecular meshworks of the patients with POAG showed more pyknotic nuclei, hyperchromatism, more disordered collagen fibers and elastic fibers. In addition, vacuolar degeneration and strait Schlemm canals could be detected. The SA-β-Gal staining exhibited that there were more positive staining in the trabecular meshworks of the patients with POAG than those of normal control (P = 0.000). 【Conclusion】 The trabecular meshworks of the patients with POAG demonstrated more severe damage in ultrastructure and more positive results in SA-β-Gal staining, which suggested apparent signs of premature senescence in the trabecular meshworks of the patients with POAG.
