摘 要: | 目的比较激光扫描共聚焦显微镜二维平面成像和三维重构成像在生物组织形态学研究中的运用价值。方法运用我院临床医学研究中心的激光扫描共聚焦显微镜对蒿草切片进行二维平面成像及三维重构成像,实验分为A组:二维平面图像组;B组:垂直叠加三维重构成像组;C组:沿X轴旋转45°三维重构成像组三个组,并对结果比较分析。结果激光扫描共聚焦显微镜不但可以获取的分辨率高和成像清晰的二维平面图像及三维重构图像,还可显示沿着X轴旋转45°的三维重构成像。结果统计显示三维重构图像的荧光值比二维平面图像的荧光值增加,另外三维图像沿X轴旋转45°后,其荧光值也增加。结论与二维平面图像比较,三维重构图像具有三维立体及含图像信息量多的特点,并可沿任意角度显示,更有利于了解对标本的空间结构关系。

APPLICATION VALUE OF LASER SCANNING CONFOCAL MICROSCOPE 3D RECONSTRUCTION IMAGING AND 2D IMAGING BASED ON THE COMPARISON ANALYSIS OF WORMWOOD SLICE |

Abstract: | Objective To compare the application value of two - dimensional imaging and three - dimension- al reconstruction imaging of laser scanning confocal microscope in a morphological study of biological tissue. Methods The two - dimensional imaging and three - dimensional reconstruction imaging of wormwood slice were performed with a laser scanning confocal microscope in our clinical research center. According to the different imaging condi- tions, the experiment was divided into three groups. Group A: two - dimensional image set; Group B : vertical stac- king of three - dimensional reconstruction imaging set; Group C : along X axis rotation angle of 45 ~ of three - dimen- sional reconstruction imaging set, and then the results were compared. Results The laser scanning confocal micro- scope not only can obtain two or three - dimensional image with high resolution and clear imaging, but also can get three - dimensional reconstruction imaging with high resolution along X axis rotation with an angle of 45 ~. The results statistics showed that the fluorescence values of the three - dimensional reconstructed image were significance higher than two - dimensional images. The fluorescence values of the three - dimensional image increased after rotating along the X axis 45 o. Conclusion Three - dimensional reconstruction of images have more image information than two - dimensional image, can get images rotated any angle along the X axis, and benefit to realize the relationship of speci- men's spatial structure. |
