大鼠力竭性游泳前后股四头肌深层肌 α-肌动蛋白基因表达的动态观察

对健康雄性大鼠力竭性游泳前后股四头肌深层肌蛋白质、ＤＮＡ、总ＲＮＡ、肌动蛋白（ＤＮａｓｅⅠ抑制法）和α－肌动蛋白ｍＲＮＡ（地高辛标记探针，Ｎｏｒｔｈｅｒｎ杂交）含量的动态观察显示：肌肉蛋白质含量游后６～１２ｈ明显升高；总ＲＮＡ含量游后即刻明显下降，１２ｈ后回升到安静时水平；游泳后Ｆ－肌动蛋白升高的趋势不明显，Ｇ－肌动蛋白下降，１２ｈ后回升，Ｔ－肌动蛋白表现出游后逐步下降，１２ｈ后出现上升的趋势；α－肌动蛋白ｍＲＮＡ的表达游后即刻略增高，６ｈ达最高，１２ｈ基本回降到安静时水平。这些变化显示了游泳运动后，大鼠骨骼肌肌动蛋白及其基因表达，以及蛋白质、ＤＮＡ和总ＲＮＡ代谢的特点。

Dynamic Changes of α Actin Gene Expression in the Profound Muscularis of M.Quadriceps Femoris of Rats after Exhausted Swimming

In this paper ,we studied dynamic changes of total protein,DNA,total RNA,T-,G-and F-actins(DN ase Ⅰ assay),and α actin mRNA(digoxigenin-labeled cDNA probe,Northern blot ) in the profound muscularis of m.quadriceps femoris of normal male SD rats before and after exhausted swimming.The results showed that total protein contents in the muscle increased from 6h to 12h after the exhausted swimming ;total RNA significantly decreasd immediately after the swimming ,and then began to increase ,and reached normal level at 12h after the swimming;Gactin levels decreased greatly at 6h and increased again at 12h after the swimming;after the swimming,T-actin contents decreased progressively and began to increase at 12h after the swimming ,α actin gene expression reached its highest level at 6h and returned to its original level at 12h after the swimming .These outcomes revealed the general patterns of changes of α-actin gene expression as well as actin ,total protein and total RNA in the skeletal muscle of rats after exhausted swimming.
