补肾固髓片对实验性变态反应性脑脊髓炎动物发病和血中IL-2、IL-6、TNF和MBP的影响

摘    要:目的:观察中药补肾固髓片对实验性变态反应性脑脊髓炎(EAE)的发病及血中IL-2、IL-6、TNF和MBP的影响。方法:本实验采用兔脊髓匀浆和完全福氏佐剂制成的抗原-佐剂乳化液诱导豚鼠EAE动物模型,应用补肾固髓片作为治疗药物,以醋酸泼尼松作为对照药物,采用放射免疫分析法测定血中IL-2、IL-6、TNF和MBP的水平。结果:大剂量补肾固髓片能明显抑制EAE发病,并且能减轻其病情,同时对血清IL-2、IL-6、TNF和MBP活性有明显抑制作用;醋酸泼尼松组、中剂量补肾固髓片组部分指标与模型组比较有统计学差异,小剂量补肾固髓片组各项指标与模型组比较无显著性差异。结论:大剂量补肾固髓片能抑制IL-2、IL-6、TNF和MBP的活性,调节免疫功能,从而抑制EAE发病,疗效与醋酸泼尼松组无显著性盖异,提示中药补肾固髓片治疗MS有临床应用价值。

Abstract:Objective:To observe the effect of Bushen Gusui Tablet (BSGS) on morbidity,and serum interleukin-2(IL-2),interleukin-6(IL-6),tumor necrosis factor (TNF) as well as myelin basic protein(MBP) of experimental allergic encephalomyelitis (EAE) in guinea pig. Methods: The EAE model of guinea pig was induced by antigen-adjuvant emusive solution making up from homogenate of rabbit spinal cord and complete Freund's adjuvant (CFA),the animals were treated with BSGS as compared with prednisone acetate,which was served as control. Radioimmunoassay was applied to determining the content of serum IL-2,IL-6,TNF and MBP. Results:High-dose BSGS could markedly reduce incidence of EAE,improve the state of illness,and meanwhile inhibit activity of IL-2,IL-6,TNF and MBP in serum. There were significant differences between prednisone acetate,moderate-dose BSGS group and model group on certain indexes,while there was no significant difference of low-dose BSGS as compared with model group. Conclusion:High-dose BSGS could inhibit activity of IL-2,IL-6,TNF and MBP in serum,regulate immunologic function,thus inhibit onset of EAE. The effect of BSGS was similar to that of prednisone acetate,which indicated BSGS could be clinically applied to treating multiple sclerosis.
