蒙古族谷胱甘肽S-转移酶GSTT1和GSTM1基因多态性

摘    要:目的 探讨内蒙古地区蒙古族谷胱甘肽S-转移酶(glutathione S-transferase GSTs EC 2.5.1.18) GSTM1和GSTT1基因多态性分布特点,为内蒙古少数民族基因型研究提供相关数据。 方法 采用内对照聚合酶链反应技术(PCR)和凝胶成像分析方法,对555例内蒙古地区蒙古族个体的GSTT1、GSTM1基因缺失型频率进行了分析。结果 GSTM1基因缺失型、GSTT1缺失型在内蒙古地区蒙古族人群中检出频率分别为55.7%和65.9%。同时具有GSTM1缺失型和GSTT1缺失型个体的 检出频率为32.2%。结论 中国蒙古族人群GSTM1、GSTT1基因呈多态性分布,与汉族及其他少数民族存在一定差异。

Abstract:Objective To explore the distribution characteristics of glutathione S-transferase T1 and M1(GSTT1,GSTM1)gene polymorphisms of Mongolian population in Inner Mongolian region and provide the relevant data for studing the genotype of ethnic minority of the Inner Mongolian.Methods Genomic DNA was extracted from peripheral blood and the fragments of GSTM1 and GSTT1 were amplified by internal control allele specific PCR in 555 volunteers who were residents in Inner Mongolian region;at the same time,the genotype of GSTT1 and GSTM1 was resolved by the dense of 2.5% agarose-gel electrophoresis.DNA fragments were bound to a dye that fluoresces orange under ultraviolet light.Meanwhile,we use the software of gel-analyzed system in computer to record and analyze the photographs.Results In this studied population,the frequencies of GSTM1 null and GSTT1 null genotype were 55.7% and 65.9% ,respectively and the frequencies of the indi- viduals with combination of GSTM1 null and GSTT1 null were 32.2%.Conclusion The deficiency frequen- cies of GSTM1 and the combination of GSTM1 null and GSTT1 null in normal Mongolian population in Inner Mongolian region approximate to normal HaM people in the north of China.But the frequency of GSTT1 null is significantly higher than normal HaM people and other reported population.
