摘    要:目的建立一种简易快速的实时荧光PCR方法(Q—PCR)并用于检测正常人群中脑膜炎奈瑟菌。方法采集成都市正常人群的咽拭子1008份,分别采用培养分离法和Q—PCR法检测Nm。绪果Q—PCR法方法的灵敏度、特异性分别为100%和100%,阴性预测值(NPV)为100.00%,阳性预测值13.09%,检出限为250du/ml体系,Q—PCR方法检出时间为3h,培养法阳性结果需时4d。Q—PCR技术检测1008份正常人群的咽拭子中NmDNA的阳性率为8.33%,培养法为1.09%。结论Q—PCR技术的灵敏度高于培养法,Q—PCR法具有灵敏、特异、经济、快速的特点,可用于疾病的快速诊断,特别适合正常人群的带菌率调查。

Detection of Neisseria meningitides in Healthy Population by Real Time Quantitative PCR

Abstract:Objective To develop a rapid test of real time quantitative PCR to detect Neisseria meningitides in healthy population. Methods A total of 1 008 throat swabs were taken in healthy population in Chengdu, Neisseria meningitides was detected by SYBR Green real-time PCR and culture/isolation assay. Results Both sensitivity and specificity of real time quantitative PCR were 100%. The negative predictive value was 100%, and the positive predictive value was 13.09%. The cut - off value of the assay was 250cfu/ml. The positive rate of Neisseria meningitides DNA by real time quantitative PCR was 8. 33% and the positive rate of Neisseria meningitides by culture/isolation was 1.09%. Conclution The real-time quantitative PCR is a rapid, sensitive and specific assay for the detection of Neisseria meningitides in healthy population, which could be a valuable method for the further understanding of the epidemiology of meningococcal infection.
