代谢组学在临床慢性肾病诊断中的应用及研究

摘    要:慢性肾病已成为一个重要的公共卫生问题,其特征表现为肾功能不全,但在疾病早期没有明显临床症状,因缺乏敏感性和特异性生物标志物使疾病早期诊断和治疗复杂化致其发病率和死亡率不断升高。代谢组学在促进临床诊断、发现疾病治疗新靶点、探索潜在疾病机制等方面显示出越来越大的潜力。慢性肾病相关代谢组学研究表明,慢性肾病与许多代谢物的失调密切相关,如氨基酸、脂质,可被用作潜在生物标记进行开发。该综述从慢性肾病临床诊断研究现状、代谢组学在慢性肾病中的应用、慢性肾病相关代谢物等方面进行总结阐述。

Abstract:Chronic kidney disease, characterized by renal insufficiency without obvious clinical symptoms in the early stages, has become a major public health problem. Because of the lack of sensitivity and specific biomarkers, the early diagnosis and treatment of chronic kidney disease become complicated, and its incidence and mortality are increasing. Metabolomics has shown potentialities in promoting clinical diagnosis, discovering therapeutic targets, and exploring underlying disease mechanisms. Metabolomics studies of chronic kidney disease have shown that chronic kidney disease is closely related to the imbalance of many metabolites, such as amino acids and lipids, which can be used as potential biomarkers. In this review, we summarize the current research of clinical diagnosis in chronic kidney disease, the application of metabolomics in chronic kidney disease, and the metabolites related to chronic kidney disease.
