摘    要:目的探讨hTERT、CMV启动子调控下TRAIL基因对乳腺癌细胞的生长抑制作用。方法脂质体法将pACCMV-EGFP-TRAIL、pAChTRET-EGFP-TRA/L转染至乳腺癌细胞MDA-MB-435,MTT法检测转染前后乳腺癌细胞的增殖变化,流式细胞术检测细胞凋亡变化。结果pACCMV-EGFP-TRAIL转染组和pAChTRET-EGFP-TRAIL转染组OD490值明显低于空白对照组和空质粒转染组,流式细胞术检测细胞凋亡率也明显增高,并且pAChTRET-EGFP-TRAIL转染组稍高于pACCMV-EGFP-TRAIL转染组。结论pAChTRET-EGFP-TRAIL、pACCMV-EGFP-TRAIL对乳腺癌细胞的生长有明显的抑制作用和诱导凋亡作用,为乳腺癌的基因治疗研究奠定基础。

Abstract:Objeetive To investigate the effect of hTERT and CMV promoter which regulated tntmor targeting TRAIL on the breast cancer line MDA-MB-435.Methods The plasmid pACCMV-EGFP-TRAIL.pAChTRET-EGFP-TRAIL was transfected into the breast cancer line MDA-MB-435 by Lipofectamine 2000. The proliferation, apoptosis of the transfeeted breast cell were measured by MTr and flow cytometry. Results Compared with the control group, the inhibitory rate of cell growth and the apotosis rate of the pAChTRET-EGFP-TRAIL and pACCMV-EGFP-TRAIL were significantly high. Conclusion The pAChTRET-EGFP-TRAIL and pACCMV-EGFP-TRAIL significantly inhibit the proliferation of breast cancer cell line, and facilitate the apoptosis of MDA-MB-435, which lays a foundation for the gene therapy of breast cancer.
