转化生长因子-β1在硬皮病皮肤组织上的表达研究

摘    要:目的研究转化生化因子-β1(TGF-β1)在硬皮病(SD)发病过程中的可能作用和相关临床意义。方法38例SD患者活检皮肤组织(SD组)和10例正常活检皮肤组织(正常组)应用免疫组织化学LsAB法测定TGF-β1,对两组TGF-β1表达含量进行比较,并对SD组内系统性硬皮病(SSc)与局限性硬皮病(LSc)、早期与中晚期的皮肤损害组织上TGF-β1表达定量进行比较,分析TGF-β1表达定量与ANA滴度、病程的相关性。结果TGF-β1阳性颗粒定位在皮肤组织细胞的胞浆内和胞膜上;在SD组皮肤组织上的TGF-β1表达量显著高于正常组(P=0.000);在SD组内TGF-β1表达量在SSc与LSc、早期与中晚期内比较无统计学意义(P>0.05);TGF-β1表达定量与ANA滴度、病程无相关性。结论TGF-β1可能参与硬皮病的整个发病过程。

Abstract:Objective To investigate the role of transforming growth factor-betal(TGF-β1) in the pathogenesis of scleroderma and the relationship between this kind of cytokine and clinical indexes of scleroderma.Methods The TGF-β1 expression in lesions of 38 patients with scleroderma and 10 normal controls were detected using immunohistochemistry LsAB assay.Results The positive signal of TGF-β1 localized in intracytoplasm and cell membrane in the dermal tissue. Compered to the control groups, the positive signal of TGF-β1 was very strong in all cutaneous tissue specimens of SD (P=0.000 ). The expression of TGF-β1 was not significant defferent between clinical indexes of systemic or localized scleroderm, at the early or middle and later stages of diseaserespectively(P>0.05). The expression of TGF-β1 had no correlation to antibody to nulear antigen(ANA) titre or course of scleroderma.Conclusion TGF-β1 may palys a role in the pathogenesis of scleroderma.
