形觉剥夺性近视豚鼠视网膜中神经生长因子及其受体的表达

摘    要:目的 探讨视网膜神经生长因子(NGF)及其受体(TrkA)在豚鼠形觉剥夺性近视(FDM)视网膜中的表达.方法 对出生7 d豚鼠以半透明眼罩遮盖右眼2个月制备FDM动物模型,左眼作为对照.遮盖前及遮盖2、4、8周后,检影验光检测双眼屈光度,A型超声测量双眼眼轴长度.免疫组织化学和逆转录一聚合酶链式反应(RT-PCR)检测视网膜NGF和TrkA的表达变化.结果 形觉剥夺使豚鼠眼轴增长,近视屈光度增加,遮盖眼与对照眼相比屈光度差异有统计学意义(P<0.01).遮盖眼NGF与TrkA免疫活性逐渐降低,NGF与TrkAmRNA表达下调,与对照组比较差异有统计学意义(P<0.01).结论 NGF和TrkA可能参与调控FDM的形成.

Abstract:ObjectiveNerve growth factor(NGF) exerts important effects by signaling commutation through surface membrane receptors,high-affinity tyrosine kinase receptors(trkA) and p75NTR between nerve tissue and non-nerve tissue.Research showed that NGF was possible to take part in the development of form deprived myopia(FDM).The purpose of this paper was to study the dynamic expression and significance of NGF and trkA on retina in form deprivation myopia of guinea pigs.MethodsFDM model was created in the right eye of 40 seven-day-old guinea pigs by wearing of translucent goggles,and the left eyes of animal models served as control group.Diopter degrees of guinea pigs in both groups were detected by retinoscopy and the eye-ball axis length was determined using ophthalmic A ultrasonography.The expression of NGF and trkA were analyzed by immunohistochemistry and RT-PCR.ResultsThe diopter degrees of FDM eyes were significantly increased in comparison with the control eyes(P<0.01).The axial length of occluded eyes was significantly longer than that of contro1 eyes(P<0.01).Hematoxylin-eosin staining showed the posterior retina of the occluded eyes was thinner than that of the control eyes.NGF and trkA positive cells were mainly distributed in the inner nuclear layer and ganglion cell layer of the retina.The immunostaining expression,the level of NGFmRNA and trkA in retina in FDM eyes were decreased than control eye(P<0.01).ConclusionNGF and trkA may be involved in the regulation of FDM.
