摘    要:目的 探讨B细胞激活因子(B cell activating factor,BAFF)在支气管哮喘(简称哮喘)发病中的作用。方法 采用酶联免疫吸附测定法检测31例健康成人(对照组)和43例哮喘急性发作期和缓解期患者治疗前后血清BAFF水平,采用实时定量-PCR法检测外周血单个核细胞(peripheral blood mononuclear cells, PBMCs) BAFF mRNA表达水平,分析二者之间的相关性。采用SPSS11.5统计学软件处理数据,计量资料采用正态性检验,非正态分布资料用中位数(最小值-最大值)表示。患者年龄用x±s表示,计量资料血清BAFF水平和BAFF mRNA以中位数(最小值-最大值)表示,应用Kruskal-Wallis法比较3组间BAFF水平的差异,若差异显著, 则采用Mann-Whitney U检验作两两比较。采用Pearson直线相关分析进行相关性分析。P<0.05为差异有统计学意义。 结果 血清BAFF水平和PBMCs BAFF mRNA表达水平在急性发作期患者高于缓解期患者和对照组,差异具有统计学意义(P<0.05),而缓解期患者与对照组相比,差异无统计学意义(P>0.05);急性发作期和缓解期哮喘患者血清BAFF水平和PBMCs BAFF mRNA表达水平呈正相关(分别r=0.456,P<0.05和r=0.523,P<0.05)。结论 血清BAFF水平和PBMCs BAFF mRNA表达水平在急性发作期哮喘患者明显增高,其动态变化与病情变化有关。

Detection and clinical significance of B cell activating factor in bronchial asthma
Abstract:Objective To explore the role of B cell activating factor(BAFF) in patients with bronchial asthma. Methods The serum levels of BAFF and the BAFF mRNA in peripheral blood mononuclear cells(PBMCs) in 31 healthy volunteers(control group) and 43 patients with the acute-outbreak period and catabatic period of asthma sufferers were measured by ELISA and real-time quantitative PCR(RT-qPCR), respectively. The data were performed by SPSS11.5 statistical package for windows. The data of the serum levels of BAFF and levels of BAFF mRNA in PBMCs were in non-normal distribution, were expressed as median(minimum-maximum), and the group comparison was made by Kruskal-Wallis and Mann-Whitney U method. The correlation between the serum levels of BAFF and the BAFF mRNA in PBMCs was tested by Pearson linear correlation analysis. A P value less than 0.05 was considered to be statistical significance. Results The serum levels of BAFF and the BAFF mRNA in PBMCs in acute-outbreak period group is higher than those in the catabatic period group and control group(both P<0.05), the difference of the catabatic period group and control group is not significant(P>0.05); the serum levels of BAFF is positively correlated with the levels of the BAFF mRNA in PBMCs both in acute-outbreak period and the catabatic period group(r=0.456, P<0.05 and r=0.523, P<0.05, respectively). Conclusion The serum levels of BAFF and the BAFF mRNA in PBMCs both increase in acute-outbreak period, their dynamic changes may contribute to the development of asthma.
