摘    要:目的 :研究血管紧张素转换酶 (ACE)插入 /缺失 (I/D)多态性的检测方法。方法 :用在 PCR反应系统中加入 5 %的二甲基亚砜 (DMSO)的改良法检测了 16 8例健康人群 ACE基因型 ,并与经典 Rigat方法进行比较。结果 :两种方法检测的 II基因型频率相同 ,但在 19例 DD基因型中 ,有 5例用 DMSO改良法检测后 ,却均为 ID基因型 ,误判率为2 6 .32 %。结论 :经典 Rigat法存在着较高率 ,改良法简便、可靠 ,适合临床实验室开展及大规模人群检测。

Abstract:Objective:To study the detecting method of ACE gene I/D polymorphism. Methods:The ACE genotypes of 168 cases were measured by classic Rigat method and an improved method by adding 5% dimethyl sulphoxide (DMSO) respectively,and the results were compared.Results:The two methods got the same II genotype frequency, but classic Rigat method had mistyping of ID genotype to DD genotype(26.32%).Conclusion:The improved method by adding 5% DMSO was a rapid and simple method for obtaining accurate and reliable result, and was suitable for clinic laboratory and mass population detected.
