妊娠高血压综合征患者血清对内皮细胞 CD226分子表达和PDGF黏附分子的研究

摘    要:目的探讨妊娠高血压综合征(PIH)患者血清对体外培养的内皮细胞表达CD226的影响,和CD226分子与PDGF表达的相关性。方法用间接免疫荧光染色结合流式细胞术分析观察妊高征患者血清对于体外培养人血管内皮细胞表达CD226的作用以及该血管内皮细胞和活化血小板黏附后CD226表达百分率的变化,用ELISA定量检测不同条件下活化血管内皮细胞分泌的PDGF,进一步探讨CD226分子和PDGF在妊高征血管内皮损伤中的作用。结果①妊高征患者血清刺激血管内皮细胞表达CD226百分率明显高于对照组(P<0·001)。②活化血管内皮细胞和活化血小板孵育后,血管内皮细胞表达CD226百分率明显降低(P<0·05)。③妊高征患者血清刺激血管内皮细胞分泌PDGF的量明显高于对照组并呈时间依赖性(P<0·001),与活化血小板作用后,分泌PDGF的量更进一步升高(P<0·005)。结论①患者体内存在某种非正常刺激因素,使血管内皮细胞敏感性增强,更易于活化。②PIH患者血清活化的血管内皮细胞与活化血小板孵育后表达CD226的百分率明显降低,提示CD226分子直接或间接参与了妊高征血管内皮细胞和血小板的黏附。③CD226与PDGF的表达有时段相关性。活化血管内皮细胞分泌PDGF增多,并与CD226表达的时间基本一致,提示其可能与CD226一起参与了妊高征形成的相关病理生理机制。

Abstract:Objective To evaluate the role of the expression of CD226 in PIH, and to investigate the correlation between the expression of CD226 and PDGF in PIH.Methods The expression?regulation and adhesion of CD226 on vascular endothelial cells incubated by sera from 10 PIH patients and 10 normal pregnant women repectively was detected by flow cytometry. The PDGF in the conditioned medium of two groups was detected by ELISA. Results (1) The positive rate of CD226 in PIH group is significantly higher than that in normal pregnant woman group(P<0.001). After the incubation with platelets, the positive rate of CD226 in PIH group is lower than before(p<0.05). (2) The PDGF expression in PIH group is much higher than that in normal pregnant woman group and is dependent on the time of stimulation(P<0.001). After the incubation with platelets, a significant increase in the release of PDGF-B is observed(P<0.005).Conclusion (1)There are some abnormal factors in PIH patients , which activate vascular endothelial cells. (2)CD226 plays an important role in the adhesion of platelets to vascular endothelial cells. (3)Both PDGF and CD226 are closely related to the progress of PIH, and take part in the pathophysiologic consequences of PIH.
