大肠癌患者外周血循环癌细胞端粒酶活性的表达及其临床意义

目的 探讨大肠癌患者外周血循环癌细胞端粒酶活性的变化与临床意义.方法 外周血经梯度离心分离单个核细胞,其中可含有循环癌细胞,再应用聚合酶链反应-酶联免疫吸附(PCR-ELISA)方法检测120例大肠癌患者外周血循环癌细胞端粒酶活性.结果 120例大肠癌患者中 65 例端粒酶活性明显升高,阳性率为54.12%,端粒酶活性的表达与大肠癌的病理类型,分化程度无明显相关性,但与淋巴结的转移明显相关 .结论 外周血循环癌细胞端粒酶活性水平与大肠癌的转移密切相关.

Expression of telomerase activity in circulating tumor cells of patients with colorectal carcinoma and its clinical significance

Objective To investigate telomerase activity in circulating tumor cells of patients with colorectal carcinoma.Methods Mononuclear cells containing tumor cells in peripheral blood of 120 patients with colorectal carcinoma was isolated by Ficoll-Hypaque gradient centrifugation. Telomerase activity of tumor cells was measured by use of polymerase chain reaction (PCR)-ELISA.Results T elomerase activity was markedly increased in 65 patients with colorectal carcinoma, the positive rate was 54.12%(65/120). There was no significant correlation between telomerase activity and pathological types or differentiation degrees of the cancer, but telomerase activity was significantly correlated with lymph node metastasis.Conclusion Telomerase activity of peripheral blood tumor cells is associated with the lymph node metastasis of colorectal carcinoma.
