摘    要:目的制备HBV基因分型芯片模板。方法通过不同基因型HBV全基因组的序列比对,选取6个碱基位点作为分型位点,应用重叠区扩增基因拼接法(SOEing法)对C型HBV片段进行点突变。结果在所选位点上成功引入点突变。结论SOEing法是一种简便引入点突变的方法,可以应用此法制备HBV基因分型检测基因所需的模板DNA。

Abstract:Objective To prepare HBV DNA fragments for HBV genotypes used as templates. Methods Six bases were chosen for genotyping by comparing different HBV genome sequences. The fragments of HBV genotype C were mutated by using gene splicing by overlap extension (gene SOEing). Results The chosen sites were successfully mutated. Conclusion Gene SOEing is a convenient way to introduce mutation,and this method can be used to prepare HBV specific sequences that can be used as templates.
