大鼠急性心肌梗死区同种异体骨髓单个核细胞移植观察

摘    要:目的:探讨同种异体骨髓单个核细胞(BM-MNCs)在急性心肌梗死区分化增殖潜能及其修复重建心肌作用,为细胞移植治疗急性心肌梗死提供新种子细胞.方法:健康雄性Wistar大鼠20只,随机分为急性心肌梗死对照组(10只)和急性心肌梗死实验组(10只).用结扎冠状动脉左前降支的方法建立大鼠心肌梗死模型,分别将培养基和制备的BM-MNCs悬液心外膜下植入梗死心肌周围,移植术后4周,观察心肌梗死区及其周边区组织形态学特点、心肌梗死面积变化.结果:移植术后4周,实验组与对照组相比心肌梗死面积缩小(P<0.01);实验组心肌梗死区内有BrdU标记阳性的BM-MNCs移植细胞存活,向心肌源性细胞分化并且诱导了大量的新生毛细血管.结论:同种异体BM-MNCs在细胞水平完成了对心肌梗死区再心肌化和再血管化过程,是治疗急性心肌梗死的理想种子细胞.

Abstract:Aim: To investigate the proliferative and differential capacities and effects on myocardial repairing and reconstruction by implanting homologous rat bone marrow mononuclear cells(BM-MNCs)in the area of acute myocardial infarction and provide a new type of seed cells for the cell-transplantation-therapy of acute myocardial infarction. Methods: A total of twenty healthy Wistar rats were divided into two groups at random:control group(AMI medium,n=10)and experimental group(AMI BM-MNCs,n=10).As soon as the descending anterior branch of left coronary artery was ligated, medium was injected into the peri-infarct area of control group and BM-MNCs labeled with BrdU were injected into the peri-infarct area of experimental group. Four weeks later, histomorphologic characteristics were observed and the areas of myocardial infarction were compared. Results: The areas of myocardial infarction were significantly smaller in experimental group(P<0.01). BM-MNCs labeled with BrdU were observed around the injected sites,differentiated into myocardium-like cell and induced angiogenesis.Conclusion: Homologous BM-MNCs are one of ideal seed cells for the cell transplantation therapy of acute myocardial infarction,which can induce cellular cardiomyoplastry and angiogenesis.
