Abstract: | Injection of Xenopus oocytes with mRNA isolated from 1-day-old rat brains leads to expression of Na+ gradient-dependent Ca2+ uptake activity. Size fractionation of the mRNA by sucrose density gradient centrifugation reveals that an mRNA fraction enriched in 14-18 S mRNA is responsible for the transport activity. Plasma membrane proteins isolated from oocytes injected with total or 14-18 S enriched fraction of rat brain mRNA contain proteins of about 70-kDa molecular mass recognized by a polyclonal antibody prepared against the purified 70-kDa rat brain Na(+)-Ca2+ exchanger. In control H2O-injected oocytes, no proteins are recognized by the anti-70-kDa antibody. |
