ATRA诱导鼠胚神经干细胞向神经元分化过程中Notch1的表达变化及意义

摘    要:目的观察全反式维甲酸(ATRA)体外诱导鼠胚神经于细胞(NSCs)分化为神经元过程中Notch1的表达变化,并探讨其机制。方法将NSCs分为两组,观察组加入终浓度为1.0μmol/L的ATRA,对照组不加ATRA。采用免疫细胞化学法检测NSCs地Notch1蛋白,采用实时荧光定量PCR法检测NSCs Notch1 mRNA。结果观察组NSCs经ATRA诱导3、5、7、9d,Notch1蛋白积分光密度值分别为539.43±13.46、488.67±13.12、437.54±13.10、434.32±13.04,Notch1 mRNA相对表达量分别为0.4122±0.0802、0.2164±0.1252、0.0221±0.1355、0.0177±0.0061;对照组分别为591.35±15.61、0.6243±0.2745,与对照组相比,P均〈0.05。结论在ATRA体外促鼠胚NSCs分化为神经元过程中,Notch1表达减少;ATRA诱导NSCs向神经元的分化与抑制Notch信号途径有关。

Abstract:Objective To explore the expression and effect of Notchl during the rat embryonic stem cells differentiating into neurons induced by all-trans-retinoic acid(ATRA) in vitro. Method The rat embryonic neural stem cells were induced differentiation with 1.0μmol/L ATRA in vitro. The expression of Notchl was respectively detected by immunocyto-chemistry,Real-time flurescent quantitive polymerase reaction and Western-Blot method, after inducing 3, 5, 7and 9 d. While in control group, the ATRA concentration was 0. Result Expression of Notchl protein and mRNA, decreased from inducing 3 d with NSCs differentiating. Conclusion The expression of Notehl decrease in the progress of NSCs differentiating into neurons in vitro. Its possible mechanism may rely on the inhibition of such signals as Notch signal pathway.
