雷公藤甲素诱导H22细胞凋亡作用研究

目的研究雷公藤甲素对H22细胞的诱导凋亡作用。方法MTT法测定雷公藤甲素对H22肿瘤细胞的抑制率,并通过细胞形态学观测及DNA凝胶电泳分析检测雷公藤甲素对H22细胞的诱导凋亡作用。结果雷公藤甲素80μg/L抑制率达41.6%,与浓度成正相关,IC50为85.79μg/L;电镜结果表明,40μg/L以上浓度可明显诱导H22细胞出现凋亡特征;DNA电泳显示明显的梯状条带。结论雷公藤甲素能诱导H22细胞凋亡,表明其具有一定的抗肝癌作用。

Study of inducing H22 cell apoptosis action of triptolide

Objective It is to study the inducing H22 cell apoptosis action of triptolide.Methods The inhibition ratio of triptolide on H22 tumor cell was determined with MTT method.The inducing H22 cell apoptosis action of triptolide was observed and detected with cell morphology and DNA gel electrophoresis analysis.Results The inhibition ratio of 80 μg/L triptolide was 41.6%.It was positively correlated with concentration and the IC50 was 85.79 μg/L.The electron microscope result manifested that the concentration of triptolide more than 40 μg/L could obviously induced H22 cell showing apoptosis characteristics.DNA electrophoresis showed obvious ladder shape strap.Conclusion Triptolide can induce H22 cell apoptosis,it shows that triptolide has certain antihepatocarcinoma effect.
